1. Properties: This product should be white crystalline powder.
2. Content determination: according to high performance liquid chromatography (Chinese Pharmacopoeia 2010 edition Appendix VI D) determination.
Chromatographic conditions: The octadecylsilane bonded silica gel was used as filler, (250*4.6mm*5.0μm); Acetonitrile: water =15:85 (pH adjusted by acetic acid =2.5) as mobile phase; The detection wavelength was 258nm. Flow rate: 0.5ml/min; The number of theoretical plates according to the HBTU
The main peak should be calculated no less than 2000.
Determination method: Take appropriate amount of this product, make the mobile phase solution containing HBTU 20μg per 1ml, as the test solution. 10 μL of the test solution was injected into the chromatograph and twice the retention time of the principal component peak was recorded.
Results: According to the area normalization method, the HBTU content should not be less than 99.0%.
3. Melting point: according to the melting point determination method (Appendix VIIC of Chinese Pharmacopoeia 2010 edition), the heating rate is 3℃/min.
The melting point of this product is not less than 200.0℃.
4. Loss on drying: take this product to 80℃ and dry under reduced pressure for 4 hours to constant weight, and the weight loss should not exceed 0.50%.
5. The clarity of the solution: take 1g of this product, add 10ml of acetonitrile, shake well, the solution should be clear and transparent.

HBTU is a coupling reagent commonly used in solid phase peptide synthesis. HBTU has also been used in standard Fmoc-based solid-phase peptide synthesis (SPPS) protocol.
HBTU, after being introduced in 1978, HBTU agent shows popularity in chemistry and industry use due to its mild activating properties. moreover, it also shows resistance against racemization. low tendency for racemization is a key requirement for peptide synthesis. especially for solid phase peptide synthesis, quantitative yields with short reaction times are crucial in order to make the synthesis of large peptides feasible.
